Nanowizard IV XP Hybrid Stage atomic force microscope (JPK-Bruker). The AFM is equipped with 100x100x15 um^3 XYZ piezos and coupled to a Hybrid Stage. The hybrid stage has extra XYZ piezos (100um each) and XY stepper motors for large range mouvements.The AFM is coupled with a Zeiss AxioObserver Z1 epi-fluorescence inverted microscope and with a top-view optical system. With the Direct Overlay software module, optical images (coming from one of the sources) can be used to define regions of interest to be scanned by the AFM.

Available imaging modalities are contact mode, Alternative Contact (AC, corresponing to Bruker's Tapping), PeakForce Tapping, Quantitative Imaging (QI) and Force Modulation mode. The miscroscope can be operated in force spectroscopy in the following modalities: standard ramping (ramp), Smart Mapping, Force Mapping (FM, corresponding to Bruker's Force Volume) and Quantitative Imaging (but force curves cannot be saved). In Smart Mapping mode only, if piezo limits are reached, the instrument automatically moves the X, Y or Z motors to recenter the sample into the piezos range.

2 sample holders are available:

• a simple holder for glass slides and large (50/60mm) Petri dishes;
• a Petri Dish Heater holder, accepting only 35mm Petri Dishes (see in the manual for a list of compatible dishes). This holder can control sample temperature by heating up to 50°C (it cannot cool samples down).

When working xith Petri dishes, place your sample close to the center of the dish.
